چکیده

A novel use of Edman degradation was made in the study of desmosineand isodesmosine-containing elastolytic peptides of bovine ligamentum nuchae elastin. The elastolysis of elastin produced peptides with the cleavage at or near the NH,-terminals of desmosine and isodesmosine cross-links. Therefore, it became possible to release single chain peptides from the carboxyl groups of these cross-links with the use of Edman degradation. This approach permitted the isolation and sequencing of seven unique peptides from bovine ligamentum nuchae elastin; the sequences of five such peptides from porcine aortic elastin were also determined. The comparison of the sequences of these peptides with the NH,terminal sequences of porcine tropoelastin tryptic peptides (FOSTER,J.A.,BRUENGER, E., GRAY, W. R., AND SANDBERG, L. B. (1973) /. Biol. Chem. 248, 2876-2879) permitted the identification of 12 possible lysine sequences which may be involved in cross-link formation. On the basis of the available data on elastin and tropoelastin, it is proposed that the tropoelastin molecule contains six pairs of lysines in the sequence -Lys-Ala-Ala-Lysand six additional pairs of lysines in the sequence -Lys-Ala-Ala-Ala-Lysand that two such pairs meet to form desmosine or isodesmosine cross-links.

Effect of adding Nanoclay (Cloisite-30B) on the Proton Conductivity of Sulfonated Polybenzimidazole

A novel sulfonated polybenzimidazole/organoclay (Cloisite-30B) (SPBI/clay) nanocomposite membranes was successfully synthesized based on aromatic diacide (1) and diaminobenzidine. Nanocomposite membranes were fabricated using 1, 4-bis (hydroxymethyl) benzene (BHMB) as cross-linker, and Cloisite-30B organoclay as the pseudo cross-linker. The cross-linked SPBI/clay nanocomposite membranes were pr...

The Effect of Structural Parameters on the Cross-Linking of Various Grades of LLDPE

Polyethylene’s application for insulating is highly advanced but due to thermoplastic properties, its application confronts constraints such as the limited 70°C temperature of the conductor. Polyethylene thermosetting procedure in which molecules are knotted and a 3D-molecular-network formed is developed to conquer the mentioned problem and to raise the applicable temperature of the i...

A structural model for desmosine cross-linked peptides.

Desmosine-enriched peptides were isolated from a thermolysin digest of bovine ligamentum nuchae elastin and a partial sequence was determined. A 'two-cross-link' model is proposed in which a second cross-link, perhaps lysinonorleucine, joins two peptide chains approx. 35 amino acid residues removed from the desmosine. Implied in this model is a certain asymmetry or directionality which places r...

Mechanistic Insights into Elastin Degradation by Pseudolysin, the Major Virulence Factor of the Opportunistic Pathogen Pseudomonas aeruginosa

Pseudolysin is the most abundant protease secreted by Pseudomonas aeruginosa and is the major extracellular virulence factor of this opportunistic human pathogen. Pseudolysin destroys human tissues by solubilizing elastin. However, the mechanisms by which pseudolysin binds to and degrades elastin remain elusive. In this study, we investigated the mechanism of action of pseudolysin on elastin bi...

Chromosomal mapping of quantitative trait loci controlling elastin content in rat aorta.

Extracellular matrix molecules such as elastin and collagens provide mechanical support to the vessel wall. In addition to its structural role, elastin is a regulator that maintains homeostasis through biologic signaling. Genetically determined minor modifications in elastin and collagen in the aorta could influence the onset and evolution of arterial pathology, such as hypertension and its com...
